This kit provides a simple and rapid method for protein quantitation based on the colorimetric Bradford assay originally described in 1976. When mixed with a protein solution, the acidic Coomassie dye reagent changes color from brown to blue in proportion to the amount of protein present. Measurements are made by comparing the color response to protein standards, typically using bovine serum albumin (BSA) or bovine gamma globulin (BGG).

The assay is compatible with most salts, solvents, buffers, thiols, reducing agents, and chelating agents found in protein samples, and can be performed in test tubes, cuvettes, or microplates.

작동 원리

Color development in Coomassie dye-based (Bradford) protein assays is associated with the presence of basic amino acids (arginine, lysine, histidine) in proteins. Van der Waals forces and hydrophobic interactions contribute to dye binding. The number of dye molecules bound is proportional to the positive charges on the protein.
Free amino acids, peptides, and low molecular weight proteins (below ~3,000 Da) do not produce color with Coomassie dye reagents.
