Target Information
SRC2 (steroid receptor coactivator 2), also known as nuclear receptor coactivator 2 (NCOA2 or TIF2), functions as a coactivator for nuclear hormone receptors.
These receptors act as conditional transcription factors that regulate gene expression involved in cell growth, development, and homeostasis.
Members of this receptor family include steroid receptors and class II nuclear receptors, which share three structural domains:
- N-terminal transcriptional activation domain
- Central DNA-binding domain
- C-terminal hormone-binding domain
Before hormone binding, steroid receptors remain inactive in a complex with heat-shock proteins.
Hormone binding induces conformational changes, leading to dissociation from inhibitory complexes, dimerization, and binding to DNA enhancer elements to modulate gene transcription.
Transcriptional coactivator proteins such as SRC2 mediate transcription initiation following enhancer binding.
