Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Xeroderma pigmentosum type G (XPG) is a human genetic disease exhibiting extreme sensitivity to sunlight. The XPG protein, a member of the flap endonuclease 1 (FEN-1) structure-specific DNA repair endonuclease family, is essential for DNA repair of solar UV-induced DNA damages. Human XPG nuclease makes the 3′ incision during nucleotide excision repair of DNA, cleaving model DNA bubble structures near the junction of unpaired DNA with a duplex region. A 29-amino acid region of human XPG (residues 981–1009) contains PCNA binding activity, with Arg992 being crucial for this interaction. Replication Protein A (RPA) binds specifically and directly to two excision repair proteins, XPA and XPG.
