Target Information

MAP4K1 (also known as HPK1) or Hematopoietic progenitor kinase 1 is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily.
MAP4K1 acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway. It diminishes T cell receptor (TCR) signaling activity and T cell proliferation by phosphorylating the adaptor protein SLP-76.
The JNKs/SAPKs play a crucial role in stress responses in mammalian cells. Understanding this pathway is important for elucidating blood cell differentiation mechanisms. HPK1 is expressed predominantly in hematopoietic progenitor cells and activates JNK1 specifically, enhancing AP-1-mediated transcriptional activity.
HPK1 binds and phosphorylates MEKK1 directly, with JNK1 activation inhibited by dominant-negative MEKK1 or MKK4/SEK mutants. Unlike PAK65, HPK1 lacks the Rac1/Cdc42-binding domain, indicating it does not interact with these small GTPases.
