Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for programmed cell death protein 1 (PD-1). PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. PD-L2 has a 2–6 times higher affinity to PD-1 compared to PD-L1.

PD-1 and its ligands function as inhibitory immune checkpoint molecules, providing negative feedback during immune homeostasis. Binding of PD-L2 or PD-L1 inhibits activation, proliferation, and cytokine secretion in T cells. Interestingly, PD-L2 can also stimulate T cell responses independently of PD-1, suggesting the presence of additional receptors. One such receptor is repulsive guidance molecule b (RGMb), which plays a role in respiratory tolerance through BMP and neogenin signaling pathways.

The human PD-L2 monomer consists of:
