Target Information
Cullin 4B is a core component of multiple cullin-RING-based E3 ubiquitin-protein ligase complexes that mediate ubiquitination and proteasomal degradation of target proteins. It functions as a scaffold protein, positioning substrates and ubiquitin-conjugating enzymes to facilitate catalysis.
CUL4B participates in polyubiquitination of CDT1, histone H2A, H3, and H4 following DNA damage, and is targeted to UV-damaged chromatin by DDB2, playing an important role in DNA repair and replication. It is required for cyclin E ubiquitination and normal G1 cell cycle progression, and regulates the mTOR pathway controlling cell growth and metabolism. CUL4B’s regulation of mTORC1 depends on 26S proteasome function and interaction with MLST8.
