Target Information
Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
FPGS has three key functions in folate homeostasis in mammals:
- Polyglutamation prevents efflux of folate cofactors from the cell.
- It enhances binding of folate cofactors to enzymes of folate interconversion and biosynthesis.
- It facilitates accumulation of folates in mitochondria required for glycine synthesis.
FPGS is widely expressed in human tumors and is associated with cell proliferation or lack of differentiation. Its integrative function contributes to the efficiency of folate antimetabolites, impacting cytotoxicity of thymidylate synthase and purine inhibitors.
