Target Information
Scn10a is a tetrodotoxin-resistant voltage-gated sodium channel alpha subunit. It mediates voltage-dependent sodium ion permeability in excitable membranes, forming sodium-selective channels that respond to voltage differences.
Scn10a is involved in neuropathic pain mechanisms. Members of the Deg/ENaC superfamily share structural similarities, including two short intracellular termini, two membrane-spanning segments, and a large extracellular loop with conserved cysteine-rich regions.
Epithelial sodium channels (ENaC) in the kidney, lung, and colon regulate sodium and fluid balance and mediate aldosterone-dependent sodium reabsorption, thereby influencing blood pressure.
Mutations in ENaC subunits can cause conditions such as Liddell’s syndrome or pseudohypoaldosteronism (PHA-1). Diseases associated with SCN10A include Episodic Pain Syndrome, Familial, 2 and Sodium Channelopathy-Related Small Fiber Neuropathy.
