Target Information
Scn5a is an integral membrane protein and tetrodotoxin-resistant voltage-gated sodium channel subunit. It is primarily expressed in cardiac muscle and responsible for the initial upstroke of the action potential in an electrocardiogram.
Defects in Scn5a cause long QT syndrome type 3 (LQT3), an autosomal dominant cardiac disease. Alternative splicing results in several transcript variants encoding different isoforms.
Voltage-gated sodium channels (VGSCs) mediate regenerative cell membrane depolarization and electrical signal conduction in nerves and muscles. Scn5a expression is also detected in lymphocytes, glia, and fibroblasts, mediating voltage-dependent sodium ion permeability of excitable membranes.
Functionally, Scn5a forms a sodium-selective channel that allows Na⁺ ions to pass according to their electrochemical gradient, contributing to the initial upstroke of the action potential. Channel inactivation is regulated by intracellular calcium levels.
Voltage-gated sodium channels (NaV) are essential for action potential initiation and propagation in excitable cells, including nerve, muscle, and neuroendocrine cells. Scn5a is also expressed at low levels in non-excitable cells, though its physiological role there remains unclear.
Diseases associated with SCN5A include Long QT Syndrome-3 and Brugada Syndrome 1.
