Product Specific Information

Description:
The monoclonal antibody 1A8-Ly6g reacts with mouse Ly-6G (Gr-1), a 25 kDa GPI protein expressed exclusively by neutrophils. It plays a role in neutrophil infiltration, recruitment, and migration, and associates with β2 integrins CD11a and CD11b. Unlike RB6-8C5, this clone binds only to Ly-6G on neutrophils and can be used for neutrophil depletion.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited with the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize nonspecific interactions.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal effect on brightness.

Target Information

Ly6G (Lymphocyte antigen 6 complex locus G6D) is part of the myeloid differentiation antigen Gr-1, along with Ly6C. It is a 21–25 kDa protein expressed by myeloid-derived cells in the bone marrow and serves as a marker for peripheral neutrophils, monocytes, and granulocytes. Expression correlates with granulocyte differentiation. Ly6G is implicated in anti-tumor responses and may modulate local immune activity via neutrophil signaling.
