Product Specific Information
For reconstitution, add 100 µL of distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating p53 stability. It is located on chromosome 12q. Akt-mediated phosphorylation at Ser166 and Ser186 enhances MDM2 interaction with p300, promoting p53 ubiquitination and degradation. Phosphorylation also prevents MDM2 binding to p19ARF, further increasing p53 degradation.
MDM2 negatively regulates p53 by inhibiting its transactivation and promoting its degradation through E3 ubiquitin ligase activity. Overexpression of MDM2 is associated with various human cancers, including soft tissue sarcomas and breast cancer.
Beyond p53 regulation, MDM2 is involved in apoptosis, cell cycle control, and tumorigenesis through interactions with proteins such as retinoblastoma 1 and ribosomal protein L5. More than 40 alternatively spliced transcript variants have been identified from tumor and normal tissues.
