Target Information
DENND3 is a guanine nucleotide exchange factor (GEF) activating RAB12. It promotes GDP-GTP exchange, converting inactive GDP-bound RAB12 into its active GTP-bound form (PubMed:20937701).
It regulates autophagy in response to starvation via RAB12 activation. Starvation triggers ULK1/2-dependent phosphorylation at Ser-472 and Ser-490, enabling recruitment of 14-3-3 adapter proteins and enhancing GEF activity toward RAB12.
DENND3 also mediates protein transport from recycling endosomes to lysosomes, influencing degradation of transferrin receptor and amino acid transporter PAT4 (PubMed:20937701).
Phosphorylation at Tyr-858 under starvation further upregulates GEF activity and initiates autophagy. [UniProt]
