The G-protein-coupled receptor (GPCR) superfamily comprises approximately 600–1,000 members and represents the largest known class of molecular targets with therapeutic value.
The nociceptin receptor is a G-protein coupled opioid receptor acting as a receptor for the endogenous neuropeptide nociceptin. Ligand binding induces conformational changes that activate G protein-mediated signaling, inhibiting adenylate cyclase and calcium channel activities. Arrestins modulate these pathways and activate MAP kinase signaling.
This receptor plays roles in modulating nociception, pain perception, and locomotor activity regulation by nociceptin.
