Target Information
FAS (Fas, CD95, APO-1)는 TNFR (tumor necrosis factor receptor) 슈퍼패밀리에 속하는 46 kDa 막관통 당단백질로, 세포 사멸 수용체로 기능합니다. FAS는 약 26 kDa의 용해형 형태도 존재합니다. FAS 자극은 FAS-associated death domains (FADD)의 응집, death-inducing signaling complex (DISC)의 형성 및 caspase 활성화를 유도합니다. 이 복합체 내의 caspase 자가절단은 apoptosis로 이어지는 하위 신호 전달을 촉발합니다. 또한 FAS는 NF-kappaB, MAPK3/ERK1, MAPK8/JNK를 활성화하여 정상 섬유아세포와 T세포의 증식 신호 전달에도 관여합니다. 최소 8개의 대체 스플라이싱 전사 변형체가 보고되었으며, 막관통 도메인을 결여한 isoform은 전체 길이 isoform에 의해 매개되는 apoptosis를 음성 조절할 수 있습니다. Fas/Fas ligand 시스템은 AIDS, 간염, 암 등 여러 인간 질환과 관련이 있으며, 항바이러스 면역 반응에도 관여하는 것으로 알려져 있습니다.
