Target Information

The origin recognition complex (ORC) is a highly conserved six-subunit protein complex essential for the initiation of DNA replication in eukaryotic cells. ORC binds specifically to origins of replication and serves as a platform for assembly of initiation factors such as Cdc6 and Mcm proteins. The ORC1 protein is the largest subunit of the complex, with levels varying during the cell cycle due to ubiquitin-mediated proteolysis after replication initiation. It is selectively phosphorylated during mitosis and interacts with MYST histone acetyltransferase 2 (MyST2/HBO1), involved in transcriptional silencing.
