Description:
The OX35 monoclonal antibody reacts with rat CD4, a co-receptor that participates in signaling through the T cell receptor by binding to MHC class II on antigen-presenting cells. CD4 is expressed in the thymus by CD4+CD8+ double-positive and CD4+ single-positive thymocytes, and by CD4+ helper T cells in the periphery. CD4 is a cell-surface membrane glycoprotein with four extracellular immunoglobulin-like domains.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended for ≤0.5 µg per test (per 100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information

CD4 is involved in the recognition of MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed in T helper cells but can also be found in monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. It associates with the T-cell receptor complex and Lck kinase to initiate T-cell activation. Additionally, CD4 may mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
