The monoclonal antibody 42D1 recognizes the CD85d protein (also known as ILT4, LIR-2). It is a member of the ILT/LIR/MIR receptor family. CD85d is a 110 kDa single transmembrane receptor with 3 Ig-like domains and a long cytoplasmic domain containing ITIMs (immunoreceptor tyrosine-based inhibitory motifs). Expression is confined to monocytes, macrophages, and dendritic cells. The ligand for CD85d is non-classical class I HLA-G, -B, and G1. Upon ligand binding, tyrosine phosphatase SHP-1 interacts with the ITIM. Interaction of HLA-G with ILT4/CD85d contributes to tolerance during fetal implantation and may play a role in organ allograft rejection.
Crosslinking of the receptor with the monoclonal antibody 42D1 inhibits calcium flux, enhances HLA-G tetramer staining, and restores the stimulating activity of tolerogenic dendritic cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Use 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.

Located at chromosomal region 19q13.4, LILRB2 is a member of the leukocyte immunoglobulin-like receptor (LIR) family. It belongs to the subfamily B class of LIR receptors containing two or four extracellular Ig domains, a transmembrane domain, and two to four cytoplasmic ITIMs. Expressed on immune cells, it binds MHC class I molecules and transduces inhibitory signals to control inflammatory responses and cytotoxicity, helping limit autoreactivity. Multiple transcript variants encoding different isoforms exist.
