Phosphoinositide 3-kinase (PI3K, phospho PI3K) binds to activated (phosphorylated) protein-Tyr kinases via its SH2 domain, acting as an adapter that mediates association of the p110 catalytic unit to the plasma membrane.
Phospho PI3K is essential for insulin-stimulated glucose uptake and glycogen synthesis in insulin-sensitive tissues. During insulin stimulation, it binds to IRS-1. Growth factors and hormones trigger this phosphorylation, coordinating cell growth, cycle entry, migration, and survival.
