Product Specific Information

The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor (60–70 kDa). This transmembrane protein binds IL-3 with low affinity alone and high affinity when associated with CD131 (common beta chain). CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (in 100 µL final volume).

Fluorophore Information:
PE-eFluor® 610 can be excited with laser lines from 488–561 nm and emits at 607 nm. Recommended detection filter: 610/20 band pass (equivalent to PE-Texas Red®). Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

IL-3Ra (Interleukin-3 receptor subunit alpha) belongs to the type I cytokine receptor family and forms a heterodimer with the common beta (CDw131) subunit. This complex transmits IL-3 signaling. IL-3Ra is overexpressed on acute myeloid leukemia (AML) blasts compared with normal hematopoietic cells, making it a potential therapeutic target.
