Target Information

STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. The STAT4 protein shares high similarity with STAT1 (52%) and STAT3 (47%). It can be tyrosine phosphorylated by Jak1 or Jak2, forming homo- and heterodimers with related STAT family members. Phosphorylated STAT4 binds the IFN-gamma activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity.

STAT4 expression is mainly found in the thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which is essential for IFN-gamma induction and Th1 cell development during immune responses against bacterial and parasitic infections. STAT4-deficient mice show impaired Th1 and enhanced Th2 development. Additionally, IFN-α/β activation of STAT4 is required for IFN-γ production during viral infection.
