Target Information

The cystatin superfamily is a well-established family of cysteine protease inhibitors.
Cystatins A and B (type 1) are mainly intracellular; cystatins C, D, E/M, F, G, S, SN, and SA are extracellular (type 2); and kininogens are type 3 cystatins found in the vasculature.
All true cystatins inhibit cysteine peptidases of the papain family (e.g., cathepsins) and some also inhibit legumain family enzymes.
Cystatin SA, S, and SN are primarily found in saliva, while cystatin S and SN are also expressed in tears, urine, and seminal fluid.
Cystatin C is expressed in the brain, thymus, ovary, epididymis, and vas deferens.
Cystatin D protects against oral proteinases, whereas cystatin E/M and F modulate cathepsin inhibition.
Fetuins, part of the cystatin superfamily, regulate osteogenesis, bone resorption, insulin and hepatocyte growth factor receptor activity, and systemic inflammation response.
High and low molecular weight kininogens inhibit thrombin- and plasmin-induced thrombocyte aggregation.
