Target Information
TRAFs are signal transducers for members of the TNFR superfamily and are critically involved in activation of MAP3K family members related to NF-kappaB and JNK activation and apoptosis.
TRAF7, a novel 670 amino acid protein, is a human homolog of mouse Traf7 and consists of an N-terminal RING finger domain, a central TRAF domain, a coiled-coil motif, and seven WD40 repeats at the C-terminus.
It exhibits E3-ubiquitin ligase activity for self-ubiquitination and specifically interacts with MEKK3 to mediate MEKK3-induced AP1 and CHOP activation. TRAF7 induces apoptosis through caspase-independent pathways and is involved in sumoylation and negative regulation of c-Myb, thereby influencing proliferation and differentiation of hematopoietic cells.
