Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. The BCL-2 family members are characterized by their ability to dimerize, modulating apoptotic activity. Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak play critical roles in cytochrome c release from mitochondria, initiating apoptosis. Overexpression of BCL-2, often due to translocation to the Ig heavy chain locus, is associated with follicular lymphoma. Neoplastic germinal centers in follicular lymphomas express high levels of BCL-2 alpha protein, while normal centers do not. Two transcript variants of BCL-2 differ at their C-terminal ends. Overexpression has been linked to cancers such as B-cell lymphoma and prostate cancer.
