Product Specific Information

The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and functions as a neutralizing antibody. Mouse IFN gamma is a 20 kDa cytokine produced by activated T, B, and NK cells with antiviral and antiparasitic properties. It acts synergistically with TNF alpha to inhibit proliferation of normal and transformed cells and modulates immune responses across various cell types expressing IFN gamma receptors. Glycosylation does not affect biological activity.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated for intracellular staining of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended use ≤0.06 µg per test (100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test. Titrate for best performance.

Fluorophore Information:
Alexa Fluor 660 emits at 690 nm and is suitable for spectral cytometers, multiplexed with APC, Alexa Fluor 647, and Alexa Fluor 700.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced by T-lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. IFN gamma exhibits species-specific activity; human IFN gamma does not cross-react with mouse. Functions include:

  • Antiviral and antitumor activity
  • Induction of MHC class I and II
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B cells

Activation occurs via IFN gamma receptor I/II and JAK-STAT signaling. Gene mutations are associated with aplastic anemia.
