Target Information

Protein C is a vitamin K-dependent plasma glycoprotein that is cleaved to its activated form by the thrombin-thrombomodulin complex. In its activated form, Protein C contains a serine protease domain and functions in degradation of the activated forms of coagulation factors V and VIII.

Protein C is produced in the liver and composed of two polypeptide chains. The 62 kDa proenzyme is activated by thrombin bound to an endothelial surface receptor, and the active enzyme (55 kDa) cleaves factor Va and VIIIa, thereby inhibiting blood coagulation.

Normal concentrations in human plasma are approximately 1–3 ng/mL for the active enzyme and about 3 µg/mL for the proenzyme due to rapid turnover. Mutations in the Protein C gene have been associated with thrombophilia due to Protein C deficiency, neonatal purpura fulminans, and recurrent venous thrombosis.
