Product Specific Information
The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins. D7 binds both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells. Non-activated splenocytes from Ly-6.2 strains (C57BL, SJL, 129, AKR, etc.) show higher D7-positive cell frequency compared to Ly-6.1 strains (BALB/c, C3H, NZB, etc.). Sca-1 expression increases upon activation regardless of haplotype.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometry on mouse bone marrow cells, ≤0.25 µg/test (100 µL final volume). Optimal titration recommended.
Super Bright 436 is excited with the violet laser line (405 nm) and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
Stem Cell Antigen-1 (Sca-1) is a member of the Ly-6 family, expressed on multipotent hematopoietic stem cells (HSC) and used as a marker for murine HSC. It is co-used with lineage depletion antibodies for HSC identification. Sca-1 is found in fetal liver, adult bone marrow, peripheral blood, and spleen, and may regulate B and T cell activation.
