This antibody is recommended for qualitative immunohistochemistry on normal and neoplastic formalin-fixed, paraffin-embedded tissues, viewed by light microscopy.
Staining requires boiling tissue in 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes.
This antibody does not cross-react with cow, dog, pig, or rat.

Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation.
It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.

CD20 is associated with lipid rafts, and its interaction with BCR (B cell antigen receptor) changes upon extracellular triggering.
After receptor ligation, CD20 and BCR colocalize and then dissociate before BCR endocytosis, while CD20 remains at the cell surface.

CD20 serves as a key therapeutic target for B-cell depletion, as it is highly expressed on B-cell malignancies and remains stable on the plasma membrane after antibody treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
