This product is diluted and supplied in a ready-to-use formulation.
A recommended positive control tissue is Hodgkin lymphoma; however, other positive control tissues may also be suitable.
The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue during manual qualitative immunohistochemistry (IHC). It is used after the primary diagnosis of tumors has been made by conventional histopathology.
CD30 is a single-chain glycoprotein synthesized as a 90 kDa precursor, processed in the Golgi complex into a mature 105/120 kDa phosphorylated glycoprotein. The CD30/Ki-1 antigen is expressed by mononuclear Hodgkin and multinucleated Reed-Sternberg cells in Hodgkin’s disease, by tumor cells of most anaplastic large cell lymphomas, and by a subset of activated T and B cells.
Ber-H2 helps distinguish large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and other lymphomas or carcinomas.
For optimal staining, pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, with washing steps in between. The resulting chromogenic reaction is observed under a light microscope to assist in differential diagnosis.
A positive tissue control must accompany each staining procedure.
This control tissue should include both positive and negative staining areas to verify staining accuracy. External positive controls should be freshly prepared and processed similarly to patient samples. Weakly positive controls are preferred for detecting subtle variations in antibody performance or reagent stability.
Negative controls (internal or external) may be used according to institutional guidelines. Internal negative control sites within tissue sections should show no specific staining, confirming the absence of background or non-specific binding. If specific staining occurs in negative controls, results with patient specimens must be considered invalid.
Target Information
CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was initially identified as a surface antigen on Hodgkin and Reed-Sternberg cells. Its ligand, CD30L (CD153), mediates various biological effects such as proliferation, activation, differentiation, and apoptosis.
CD30 plays a crucial role in the pathophysiology of Hodgkin’s disease and CD30+ lymphomas, and acts as a costimulatory molecule in thymic negative selection. It is also expressed in some non-Hodgkin lymphomas, virus-infected T/B cells, and activated normal lymphocytes. Soluble CD30 (sCD30) serves as a marker reflecting Th2 immune responses.
CD30 interacts with TRAF2 and TRAF5 to activate NF-κB signaling and positively regulates apoptosis, limiting autoreactive CD8 T cell proliferation. Two alternatively spliced transcript variants exist, encoding distinct isoforms.
CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
