This antibody is diluted and ready to use.
Recommended positive control tissue: hepatocellular carcinoma (HCC).

The antibody is intended for professional laboratory use in detecting Glypican-3 (GPC3) in formalin-fixed, paraffin-embedded tissue via manual qualitative IHC testing. It is used after primary diagnosis of tumor by conventional histopathology.

Glypican-3 (GPC3) is a heparan sulfate proteoglycan anchored to the cell surface. The 1G12 monoclonal antibody has been used to assess GPC3 expression in malignant and nonmalignant liver tissues.
Studies indicate GPC3 is expressed in most hepatocellular carcinomas but not in normal liver or benign hepatic lesions. Serum GPC3 levels are elevated in many HCC patients, making it a useful marker for differentiating benign and malignant liver diseases.

Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate. The resulting color reaction is observed under a light microscope for diagnostic interpretation.

A positive tissue control should be included in every staining procedure.
Positive controls may contain both positive and negative staining cells and serve as internal quality checks.
External control tissues should be freshly prepared and embedded similarly to patient samples, ideally showing weak positive staining to detect subtle changes in antibody sensitivity or reagent degradation.

Negative control tissues (internal or external) should demonstrate absence of specific staining and help verify non-specific background staining. If positive staining occurs in negative controls, patient sample results must be considered invalid.


Target Information

GPC3 is a cell-surface proteoglycan bearing heparan sulfate. It may modulate growth in mesodermal tissues and regulate IGF2 receptor interactions.
Glypican family members are anchored to the membrane via GPI linkage and are involved in cell division and growth regulation.
Deletion mutations in GPC3 cause Simpson-Golabi-Behmel syndrome (SGBS), characterized by pre- and postnatal overgrowth with visceral and skeletal anomalies.
