• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein (40–43 kDa) expressed in various tumors and normal cells, particularly lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells. Podoplanin functions as a regulator of lymphatic endothelium and may help maintain podocyte morphology.

Podoplanin acts as a ligand for CLEC-2, and its expression correlates with tumor invasiveness and metastatic potential. It is involved in cell migration, metastasis formation, and tumor invasion. Expression can be induced through Src activation of Cas, and it has been detected on a subset of Th17 cells and CD4+ PBMC populations. Dysfunction of Podoplanin is associated with diseases such as lymphangioma and subglottis neoplasm. It is also proposed as a marker for lung injury.
