PA1-913은 Western blot, immunofluorescence, immunocytochemistry, immunohistochemistry (paraffin), immunoprecipitation 등 다양한 실험에 성공적으로 사용되었습니다.
Western blot에서 약 55 kDa의 단백질을 검출하며, 이는 개 심근 추출물의 calsequestrin에 해당합니다. 추가적으로 97 kDa 부근의 밴드가 관찰될 수 있으며, 이는 calsequestrin 유사 단백질로 보고된 바 있습니다.
PA1-913의 항원은 정제된 개 심근 calsequestrin입니다.
Target Information
근형질세망(Sarcoplasmic Reticulum, SR)은 심근 및 골격근 세포 내 칼슘 수준을 유지하기 위해 칼슘을 저장하고 방출하는 역할을 합니다. SR 내강의 주요 칼슘 결합 단백질 중 하나는 calsequestrin으로, 근세포의 이완 시 칼슘을 저장하고, 수축 시 방출될 수 있도록 조절합니다.
이 단백질은 높은 산성도를 가지며, 큰 용량과 중간~낮은 친화도로 칼슘을 결합합니다. 주로 말단 시스테르나의 접합면 근처에 위치하여, ryanodine receptor를 통해 세포질로 칼슘이 방출되는 과정을 조절합니다.
