Store vial at -20°C prior to opening. Aliquot and refreeze at -20°C or below for long-term storage. Avoid repeated freeze/thaw cycles. Centrifuge if not clear after standing at room temperature. Stable for several weeks at 4°C as an undiluted liquid. Dilute only before immediate use.

This IgG preparation of whole rabbit serum (purified by DEAE fractionation) is primarily directed against mature 17 kDa human IL-1β and is useful in detecting IL-1β in various assays. It also detects primate IL-1β with similar dilutions but does not recognize human IL-1α.
In ELISA and other immunoassays, it recognizes ~10% of non-denatured precursor (31 kDa) IL-1β but primarily detects the mature 17 kDa molecule.
In immunoblots, denatured 31 kDa precursor IL-1β appears as a 35 kDa band due to unfolding, exposing epitopes not accessible in the native state. The antibody detects both the 17 kDa mature band and the 35 kDa precursor band depending on sample type.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is synthesized as a 31 kDa inactive pro-form and cleaved into an active 17 kDa form by caspase-1 following inflammasome activation.
IL-1β lacks a signal peptide for classical secretion and is released via an alternative pathway. It signals through IL-1RI and IL-1RII receptors, shared with IL-1α, and its activity is regulated by IL-1 Receptor Antagonist (IL-1RA).
IL-1β plays a key role in innate immune responses, promoting cytokine production and acute-phase reactions. Elevated IL-1β levels are linked to chronic inflammatory diseases, and IL-1β neutralizing antibodies have therapeutic potential.
