Target Information

Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one utilizing NAD(+) and the other NADP(+) as electron acceptors.
Five isocitrate dehydrogenases have been reported: three NAD(+)-dependent forms localized to the mitochondrial matrix, and two NADP(+)-dependent forms — one mitochondrial and one predominantly cytosolic.
The encoded protein is the NADP(+)-dependent isocitrate dehydrogenase found in the cytoplasm and peroxisomes, containing the PTS-1 peroxisomal targeting signal sequence.
Its presence in peroxisomes suggests roles in NADPH regeneration for intraperoxisomal reductions (e.g., conversion of 2,4-dienoyl-CoAs to 3-enoyl-CoAs) and in peroxisomal reactions consuming 2-oxoglutarate, such as alpha-hydroxylation of phytanic acid.
The cytoplasmic enzyme plays a key role in cytoplasmic NADPH production.
