Product Specific Information

For reconstitution, add 100 µL distilled water to achieve a final concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

Modification of target proteins by ubiquitin participates in diverse biological functions. Proteins destined for degradation via the 26S proteasome are coupled to multiple ubiquitin copies. Attachment of ubiquitin or related molecules can influence subcellular distribution or protein activity.
Deubiquitination, the removal of ubiquitin, is catalyzed by deubiquitinating enzymes (DUBs), grouped into four families:

  1. Ubiquitin C-terminal hydrolases
  2. Ubiquitin-specific processing proteases (USPs)
  3. OTU-domain ubiquitin-aldehyde-binding proteins
  4. Jab1/Pad1/MPN-domain-containing metallo-enzymes

USPs are the most widespread DUBs, releasing ubiquitin from conjugated proteins. They possess conserved catalytic Cys and His boxes, with variable terminal extensions for substrate recognition and localization.
