세포 내 위치

Cytoplasm, cytoskeleton, cilium basal body, cell junction (tight junction), microtubule organizing center (centrosome, centriole), cytoplasmic vesicle (autophagosome), Golgi apparatus, nucleus, and spindle.
Co-localizes to cytoplasm only in presence of ESRRA (PubMed:21190936). Translocates to nucleus upon activation (PubMed:20638370). Detected at spindle during meiotic stages (By similarity).

Background

  • Catalytic activity: ATP + a protein = ADP + a phosphoprotein.
  • Domain: N-terminal (1–20) region necessary for ubiquitination and proteasomal degradation.
  • TXY motif: Contains Thr and Tyr residues whose phosphorylation activates MAP kinases.
  • Enzyme regulation: Activated by Thr and Tyr phosphorylation; inhibited by dual specificity phosphatases (e.g., DUSP1).
  • Function: Phosphorylates MBP in vitro.
  • PTM: Dually phosphorylated on Thr-175 and Tyr-177; autophosphorylated in vitro; ubiquitinated for regulation and turnover (possibly by SCF E3 ligase).
  • Similarity: Member of protein kinase superfamily, CMGC Ser/Thr protein kinase family, MAP kinase subfamily; contains one protein kinase domain.
  • Subunit: Interacts with CSK/c-Src, ABL1, RET, and TGFB1I1.
  • Tissue specificity: Widely expressed, highest in lung and kidney.
