Background
This gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. The ErbB-3 protein possesses a neuregulin binding domain but lacks an active kinase domain, enabling ligand binding without signal transduction via phosphorylation. It forms heterodimers with other EGFR family members that have kinase activity, leading to activation of pathways involved in cell proliferation and differentiation. Amplification or overexpression of this gene has been observed in various cancers, including prostate, bladder, and breast tumors. Multiple splice variants exist, including a secreted isoform that modulates membrane receptor activity.
