Background

This gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. This membrane-bound protein has a neuregulin binding domain but lacks an active kinase domain. It can bind the ligand but cannot convey the signal through phosphorylation. However, it forms heterodimers with other EGFR family members possessing kinase activity, leading to activation of pathways responsible for cell proliferation and differentiation. Amplification or overexpression of this gene has been reported in various cancers, including prostate, bladder, and breast tumors. Alternative splicing generates isoforms, one of which is secreted and modulates the activity of the membrane-bound form.
