Target Information

Ankyrins are a family of proteins that link integral membrane proteins to the underlying spectrin–actin cytoskeleton. They play key roles in cell motility, activation, proliferation, contact, and maintenance of specialized membrane domains. Multiple isoforms of ankyrin are expressed in a tissue-specific and developmentally regulated manner.

Most ankyrins consist of three structural domains:

1. Amino-terminal domain with multiple ankyrin repeats
2. Central region with a conserved spectrin-binding domain
3. Carboxy-terminal regulatory domain (least conserved and variable)

Ankyrin 1, the prototype of this family, was first identified in erythrocytes and later found in brain and muscle. Mutations in erythrocytic Ankyrin 1 are associated with hereditary spherocytosis. Various isoforms arise from alternative splicing and alternate promoter usage, including truncated muscle-specific forms.
