Target Information
Plasma prekallikrein is a glycoprotein involved in blood coagulation, fibrinolysis, kinin generation, and inflammation. It is synthesized in the liver and secreted into the blood as a single polypeptide chain.
Plasma prekallikrein is converted to plasma kallikrein by factor XIIa via cleavage of an internal Arg-Ile bond, resulting in a heavy and light chain linked by a disulfide bond.
- The heavy chain contains four tandem repeats with apple domains, essential for surface-dependent pro-coagulant activity.
- The light chain contains the active catalytic domain, homologous to trypsin-like serine proteases.
Deficiency of plasma prekallikrein leads to prolonged activated partial thromboplastin time.
