Target Information

SHOC2 protein participates in protein binding and transferase activity within the fibroblast growth factor receptor signaling pathway and Ras protein signal transduction.
It is a widely expressed protein composed almost entirely of leucine-rich repeats (LRR) with a lysine-rich sequence at the amino terminus and localized in the cytoplasm.
SHOC2 acts as a positive modulator of the RAS-MAPK signaling cascade, mediated by EGL-15, LET-23, and LET-60. Together with protein phosphatase 1c (PP1c), SHOC2 forms an M-Ras effector complex essential for MAPK pathway activation.
In tumor cells with Ras mutations, inhibition of SHOC2 suppresses MAPK but not PI3K activity, making the SHOC2-PP1c holoenzyme a potential therapeutic target.
Aberrant N-myristoylation of SHOC2 has been linked to Noonan-like syndrome with loose anagen hair.
