Product Specific Information

The mIDO-48 monoclonal antibody reacts with mouse indoleamine 2,3-dioxygenase 1 (IDO1).
IDO1 is an intracellular enzyme expressed by dendritic cells, IFN-γ-activated macrophages, epithelial and endothelial cells, and tumor cells. It catalyzes the conversion of tryptophan into kynurenine metabolites, leading to suppression of T cell proliferation and induction of regulatory T cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Intracellular staining of mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

  • Recommended use ≤ 0.125 µg/test (100 µL final volume)
  • Cell number: 10⁵–10⁸ cells/test (empirically determined)
  • Optimize antibody titration for best performance

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by a 488 nm blue laser.
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility.

Light Sensitivity:
Protect vial and stained samples from light; tandem dye is sensitive to photo-induced oxidation.

Target Information

IDO1 (indoleamine 2,3-dioxygenase 1) is a heme-containing intracellular enzyme that catalyzes the oxidative cleavage of the indole ring in tryptophan, serotonin, and melatonin, producing kynurenine metabolites.
IDO1 is expressed in multiple tissues and immune cells (macrophages, DCs) and is induced by interferons (IFNs).
IDO1 plays a critical role in antimicrobial defense and immune regulation in infection, pregnancy, autoimmunity, transplantation, and cancer.
