Target Information
The accumulation of unfolded proteins in the endoplasmic reticulum (ER) triggers the ER stress response. This includes inhibition of translation to prevent further accumulation of unfolded proteins, increased expression of proteins involved in folding (unfolded protein response, UPR), and destruction of misfolded proteins via the ER-associated protein degradation (ERAD) system.
HERPUD1 may play a role in both UPR and ERAD. Its expression is induced by ER stress and contains an ER stress response element in its promoter. The protein has an N-terminal ubiquitin-like domain, may interact with the ERAD system, and has been shown to interact with presenilin proteins, increasing amyloid-beta levels upon overexpression.
Alternative splicing produces multiple transcript variants, some encoding different isoforms.
