SYNE1과 SYNE2 유전자는 type II integral membrane 단백질 계열에 속하며, 다양한 조직에서 발현됩니다. 대체 전사 개시 및 스플라이싱에 의해 여러 isoform이 생성되며, 일부는 N-말단이 절단된 형태로 존재합니다.
이 단백질들은 nuclear envelope spectrin repeat (nesprin) 단백질로, 다수의 spectrin repeat, 이중 핵 내 위치 신호, 그리고 보존된 C-말단 단일 transmembrane domain을 포함합니다.
EGFP-fusion construct를 이용한 일시적 발현 실험에서 핵막에 국한되어 발현됨이 확인되었으며, 이는 perinuclear localization에 필수적인 C-말단 TM domain에 의해 매개됩니다.
Nesprin-1은 평활근 및 골격근에서 발달 단계별로 조절되며, C2Cl2 myoblast 분화 과정에서 핵막에서 핵 및 세포질로 재배치됩니다.
