Target Information
NALCN (Sodium Leak Channel, Non-selective)
NALCN is a voltage-independent, cation-nonselective channel permeable to sodium, potassium, and calcium ions. It regulates the resting membrane potential and neuronal excitability. Neuropeptides such as neurotensin and substance P activate NALCN via SRC family kinases.
NALCN activity is modulated by GPCRs, essential for normal respiratory rhythm, neonatal survival, and systemic osmoregulation by controlling serum sodium concentration. It contributes to depolarization induced by substance P and regulates intestinal pacemaker activity in interstitial cells of Cajal.
Additionally, NALCN is crucial for the spontaneous firing and excitability modulation of substantia nigra pars reticulata (SNr) neurons. Diseases associated with NALCN include congenital contractures of the limbs and face, hypotonia, developmental delay, and infantile psychomotor retardation with characteristic facies.
