Target Information
Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates specific prolines in the oxygen-dependent degradation domains (NODD and CODD) of HIF1A and HIF2A, leading to their proteasomal degradation under normoxia. Under hypoxia, hydroxylation is reduced, allowing HIFs to accumulate and activate hypoxia-responsive genes. EGLN3 (PHD3) also regulates apoptosis in cardiomyocytes and neurons, glycolysis through PKM hydroxylation, and DNA damage response via TELO2 hydroxylation.
