Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases, and participates in receptor downregulation via ubiquitin-mediated removal.
CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues on N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia (AML). It is found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells. CD33 expression on peripheral monocytes makes it a useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
