Target Information
UBE2O is an E2/E3 hybrid ubiquitin-protein ligase with both E2 and E3 ligase activities. It mediates monoubiquitination of target proteins and regulates multiple signaling pathways:
- Negatively regulates TRAF6-mediated NF-κB activation independently of its E2 activity.
- Acts as a positive regulator of BMP7 signaling by mediating monoubiquitination of SMAD6, influencing adipogenesis.
- Mediates monoubiquitination of the nuclear localization signal (NLS) of BAP1 and other chromatin-associated proteins (INO80, CXXC1), affecting their subcellular localization.
- Regulates retrograde transport by assisting the TRIM27:MAGEL2 E3 ligase complex to ubiquitinate WASHC1, promoting endosomal F-actin assembly.
