Target Information

Scn5a is an integral membrane protein and tetrodotoxin-resistant voltage-gated sodium channel subunit. It is found primarily in cardiac muscle and is responsible for the initial upstroke of the action potential in an electrocardiogram.
Defects in Scn5a cause long QT syndrome type 3 (LQT3), an autosomal dominant cardiac disease. Alternative splicing results in several transcript variants encoding different isoforms.
Voltage-gated sodium channels (VGSCs) mediate regenerative cell membrane depolarization and electrical conduction in nerves and muscles. Expression of Scn5a is also detected in lymphocytes, glia, and fibroblasts.
Scn5a mediates voltage-dependent sodium ion permeability of excitable membranes, forming sodium-selective channels that allow Na⁺ ions to pass according to their electrochemical gradient.
Functionally, Scn5a is responsible for the initial upstroke of the action potential, and its inactivation is regulated by intracellular calcium levels.
Diseases associated with SCN5A include Long QT Syndrome-3 and Brugada Syndrome 1.
