Target Information
The differentiation and activation of monocytes (MO) and monocytic cells is modulated by 1α,25-dihydroxyvitamin D3 (Vitamin D3). Using mRNA Differential Display technology, Fritsche J et al. identified a gene induced in freshly isolated human blood MO cultured with Vitamin D3. This cDNA encodes a putative 16.5 kDa protein structurally homologous to yeast MMS2p and human CROC-1, defining a conserved Ubc-like protein family. The protein, called Ubiquitin E2 variant (UBE2V2) or MMS2, is essential for RAD6/RAD18-dependent postreplicative DNA repair and K63-linked polyubiquitin chain assembly. UEV/Ubc complexes play a key role in DNA repair and IκBα kinase activation. Crystal structure analysis confirms that the Mms2/Ubc13 heterodimer is necessary for DNA repair.
