Target Information

Serine/threonine-protein kinase (SIK1) is involved in various biological processes such as cell cycle regulation, gluconeogenesis, lipogenesis, muscle growth, differentiation, and tumor suppression.
It phosphorylates HDAC4, HDAC5, PPME1, SREBF1, and CRTC1/TORC1, and inhibits CREB activity by phosphorylating TORCs (CRTC2/TORC2, CRTC3/TORC3) in response to cAMP signaling.
SIK1 acts as a tumor suppressor, mediating p53-dependent anoikis and suppressing metastasis. It also participates in sodium-sensing signaling networks through phosphorylation of PPME1 and regulation of ATP1A1.
In muscle cells, SIK1 promotes MEF2 target gene expression by inhibiting HDAC4 and HDAC5. It regulates hepatic gluconeogenesis and lipogenesis by repressing CREB coactivators and SREBF1, and contributes to circadian rhythm regulation via CRTC1/TORC1.
SIK2 phosphorylates IRS1 (Ser-794) in insulin-stimulated adipocytes, modulating insulin signaling efficiency.
SIK3 positively regulates mTOR signaling by promoting DEPTOR degradation, influencing chondrocyte differentiation during skeletogenesis, and negatively regulating cAMP signaling through TORCs.
